A recent scientific study published in the British Medical Journal found a connection between prolonged use of the popular injectable birth control drug Depo-Provera and the development of a type of brain tumor known as a meningioma. What is Depo-Provera?

Depo-Provera, or depot medroxyprogesterone acetate, is an injectable birth control method for women that is typically injected into the arm or buttocks every three months. The drug contains progestin, a synthetic compound that mimics the effects of the hormone progesterone, which suppresses ovulation and thickens cervical mucus to prevent pregnancy. However, scientists believe that Depo-Provera’s link to meningioma arises from its high levels of synthetic progestin.

Depo-Provera is 96 percent effective in preventing pregnancy, and many women choose this form of birth control for long-term effective protection that they don’t need to take every day.

Potential Risks and Side Effects of Depo-Provera

In March 2024, the British Medical Journal published a study analyzing data from the French National Health Data System. The analysis showed an excess risk of intracranial meningioma with the use of medroxyprogesterone acetate—the active ingredient in Depo-Provera—for over a year. 

Meningiomas account for 40 percent of primary tumors of the central nervous system, growing slowly to compress adjacent brain tissue. Three risk factors make people more susceptible to developing a meningioma: age, being female, and prolonged exposure to medroxyprogesterone acetate.

The Depo-Provera label does not mention meningiomas, brain tumors, or severe risks of prolonged use. The label does state that prolonged use can cause osteoporosis, a possible increased risk of breast cancer, blood clots, strokes, and several other conditions. Despite these risks, the label declares that patients should not use Depo-Provera for over two years “unless you cannot use other birth control methods.”
